Although I don’t have many recs since I haven’t had the time to read any new fics but I hope these maybe fit what you’re looking for :) also sorry for the late reply.
On air : they’re radio hosts and their listeners are convinced that they’re dating. It’s a really good fic and I wish it was longer ngl.
Plot twist : it’s based on the ‘i thought i was homophobic but turns out im in love with my roommate’ reddit post..yeah.. Dean’s an idiot and we love that about him.
Stay with me, sweetheart : i love this fic so much. It’s Firefighter!Dean and Teacher!Cas. And Cas gets into an accident and Dean helps him during and after. Basically strangers to friends to lovers. Also Cas is a dad.
r/Relationships : i really liked this fic. It’s strangers to friends to lovers. It’s also them kind of having an aha moment with their sexualities.
The barista and the bookshop : this one is is on my ‘to read’ list so I can’t give a full opinion yet. But I’ve seen it recommended a lot which is how I came across it.
Mixtape for My Teenage Crush : this one is also on my ‘to read’ list. (Can you tell I’ve been busy and haven’t had the time to read fics that are longer?)
What's the Problem Baby? : They’re idiots in love. It’s only 2 chapters but it makes me want to grab them and shake them for being so dumb.
Vacation, All I Ever Wanted (Was You) : this one was so good and so cute. Dean is a workaholic (dean smith type) and Cas loves him and wants only the best for him, so obviously he takes him on a trip that keeps going wrong…

Sometimes I use these deep dive recs to highlight longform journalism or well-researched nonfiction, which might make this seem like a classy and reputable establishment, but it's important to remember that this is tumblr, and none of us are free from sin. This week's rec is the Ride or Die podcast, which hosts Pru and Waldorph describe as a death march through CW's Supernatural, episode by episode.
Ride or Die also does double duty as an ethnographical case study of The Destiel Phenomenon for anyone who missed seeing it develop live. There's little I like more than close reads of media with genuinely high highs and infamously low lows, and I don't always agree with Pru and Waldorph about which moments are which, but by the time I had formed strong opinions of my own it was already too late.

One of my favorite fics of all time is Evangelist by @valleydean. It's really enemies to friends to lovers but the enemies part becomes friends pretty quickly because they are deeply obsessed with each other.
One of the classics if you haven't read it is To Build a Home. Childhood friends who broke up and find themselves back in each other’s life. It is very angsty but they have a lifelong friendship so you get a lot of childhood and teenage friendship moments.
If I may self recommend, I just wrote Can't Fight the Moonlight in which they are in this post canon partnership and both want to add sexual intimacy but the partnership is soft so you get some really fun platonic intimacy. It's not AU, obviously, but may have something of what you want.
Seraph by @dothraki-shieldmaiden is a superhero au where they are friends and roommates to lovers. It has great world building.
More than Kisses by @friendofcarlotta is a friends to lovers epistolary romance. Dean and Cas are pen pals and they share an absolutely gorgeous emotional intimacy.
Tiamatv has a few that fit the bill. I'm gonna mention Marchen because I love the world built in that one. Dean Winchester is a prince locked in a tower.

"A Midterms Night's Dream" by Englandwouldfall is an au where they're roommates (oh my god they were rommates).
"In Some Sacred Place" by schmerzerling is a good one where they're friends throughout most of the fic, but it's kind of depressing overall

I don't know that I have any specifically best friends to lovers fic recs because imo every destiel fic is a best friends to lovers & also I'm not a huge AU person BUT.... if you'd still like some recs here are some of my favorites:
spirit of the west by teen-dean (everyone should read this 90's horse girl dean au where he befriends cas, a local veterinarian)
I carry your heart (I carry it in my heart) by unicornpoe (I am a sucker for all "cas using dean as a vessel" fics)
Après by imogenbynight (surely everyone has read this? dean and cas in paris what more can you ask for)
under the midnight sun by northernsparrow (dean is the caretaker of a research station in the arctic and cas is a mysterious bird researcher) (I also rec you can keep holding on by this author but be warned that it is very grief-driven - but it is an in-depth study of the way they care for each other)
though the course may change by imogenbynight (fake dating AU at a couple's retreat - another one that everyone has probably read but is so worth mentioning in case you haven't)
there's no cell service in the afterlife by screamsintothesun (cas gets back from the empty and his phone blows up with dean's voicemails & texts)
Gold in the Edges of Our Vision by SewingNatural (dean and cas eat peaches together on a hot summer evening. The writing style on this one is really really lovely)

Hi, I'm not sure I can help much. I quickly went through the list of fics I've read and tbah is definitely the one that fits the friends to lovers best. I found a couple others that do have some kind of friends to lovers as well. (I always don't know if it's friends to lovers if they have a crush from the start but still are friends for a while etc. so I'm not sure how much it applies to them.)

Any Little Heartbreak - fwb to partners hospital AU, so so good i literally have quotes from this on sticky notes on my wall
What I Need - i love hospital AUs and i think this is my favorite. cas is a surgeon and dean is the only surgery tech he's willing to work with.
