Using Transposases for Genetic Engineering

Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) and CRISPR-associated proteins (Cas) have been repurposed from the adaptive immune system of bacteria as tools to enable genetic engineering. However, Cas9 and Cas12 systems which have been widely employed for genome editing require host cell repair machinery, which can depend on active replication of the cells. As a result, it has largely been limited to nucleotide substitutions. 

SBGrid member Feng Zhang and other researchers have been working to use a CRISPR-associated transposase (CAST) in order to harness DNA insertion mechanisms. The researchers investigated a CAST from Scytonema hofmanni and characterized this system consisting of Tn7-like transposase subunits and Cas12k. They investigated its mechanism of RNA-guided DNA transposition and report that the DNA insertions are directed to 60-66 basepairs downstream of the protospacer, without relying on the Cas nuclease activity or endogenous homologous recombination DNA repair pathways in the target cell.

While over-expression of the construct can lead to less specific insertions, particularly into highly expressed genes, it is believed that CAST systems identified in different microbes could exhibit different properties, such as target preference and activity. The therapeutic implications of such systems may include the insertion of corrected exons into an intron before mutated exons to restore gene function.

No, it isn’t stock - but modified Mk1 Honda Preludes typically answer the main criticisms that most people had of the car when it was new. Mainly, that it was not all that sporty - straddling the line between a coupe and an economy car; and that its styling was too sedate. The S-series sports cars had been gone for nearly 7 years by the time development started on the Prelude. The S-cars were very well liked, but built in tiny numbers and two-seat cars had limited appeal in Japan the Prelude would be high-volume territory - a coupe aimed at a wide cross-section of small sedan-related coupes - Celicas, Mantas, Sunbirds, etc. This jack of all trades debuted in Japan in November, 1978. At the time, Honda only had two car lines, the Civic and the Accord. The entire company had been redefined by these cars and the older Kei models (except the TN7 truck) had been shelved in 1974. Any car it developed had to have as much potential in Japan as elsewhere, and coupes were popular. The Prelude would leverage existing components to keep costs down, but it got its own chassis courtesy of chief engineer Hiroshi Kizawa. It was broadly similar in many ways to the Accord and Civic, but as Kizawa and his team also developed those cars, that wasn’t surprising. As with the Civic and Accord, it was also styled under Honda veteran Shinya Iwakura with, as usual, heavy input from Soichiro Honda himself. It was a premium product for Honda in Japan (sold at the new Honda Verno channel along with the new Honda Quint - ancestor to the Integra), which was reflected in equipment levels, like an optional moonroof and Connolly leather. A tiny back seat kept things tax friendly in Japan but was useless for real adults. In Japan it came with two engines - a 1.6L “EL” and the CVCC 1.8L “EK,” but only the latter came stateside, with 72 hp. In the USA it was marketed as a smaller, higher-quality version of the personal/luxe coupe idea. It was a sharp handler with a tight suspension and it was built to very high standards (though, like all Japanese cars of the time, it rusted), but clearly not a sports car, particularly with a 3sp auto.
