Gunslinger is throw away answer, because of course Ruby is going to be a gunslinger. So, we have a werewolf battle nun Ruby, with rainbow magic powers, (magical girl), and sacred weapons, (vampire hunting).
I'm not picking the one with the most votes, but going to work out a way to get everything picked to work together.

It's close, Werewolf is leading, and I can do more with it.
So, it would be a werewolf that also gains a magical transformation into a magical girl.
For modern fantasy Ruby, since it was werewolf then magical girl in second have you considered werewolf with a magical girl transformation to change forms?
I was honestly thinking about that, and it would be fun and hilarious. Especially if combined with Sailor Moon.
But now it's Werewolf and Wolf Faunus, with Magical Girl barely having any more votes than Battle Nun.
Maybe have a second poll that combines the options.

The Protagonist has to have an understanding of the RPG mechanics, but ONLY the Protagonist does. People have to be able to Level Up to get stronger, (unless they are already max level). Any anime that has Status Plates, or the like, is excluded.
Anyone not ridiculously OP is excluded for their own safety.

Blood Triangle: Blood Angel, Space Wolf, SoB love triangle. Ends with an Eldar woman that asks her about it, and it turns out the SoB was too awestruck to even consider that possibility.
My Pet Xenos: Astartes ends up winning a Dark Eldar slave girl. She spends the whole story trying to seduce him, while he honestly doesn't know what to do with her. Plot requires he keep her alive until the denouement.
Astartes enters slapstick harem shenanigans, and does everything he can to not get with the crazy women around him.
The more you read it, the more it's obviously a Grim and Evil parody.
A Midsummer Night's Dream staring Lion El'Jonson. Ends up with him recruiting Exocite Eldar Dragon/Dino riders to his cause.
Ultramarine trying to keep Ultramar peaceful and stable, despite the galaxy doing everything it can to stop this. Obviously a buddy cop movie with as the bad cop.
A World Eater causes a Blood Angel to bleed, and the Blood Angels going on a roaring rampage of revenge to kill him.
Roboute Guilliman tries to do everything he can to pretend that Yvraine is not a Xenos. Obvious I Dream of Jeannie parody.
