i know for the fact that the stems from microscopic streptomycete bacteria in the soil produce a compound called geosmin which results in petrichor, the heavenly smell of the earth after the first rain and the reason why i remember this is because i find it similar to how it feels when there's you running around me spreading love - your love has an aroma my nose refuses to forget. when i see me out on the road struggling to find a taxi to go home, my eyes glance down on someone dancing in harmony with the thunder with innocence raining on them, my olfactory sensory neurons become nostalgic and make me remember your scent, but i break free from the thoughts and sit in the taxi. and i go home, change my clothes and sit in the balcony enjoying a cup of tea when my mom comes and says, "oh this beautiful fragrance of the earth, can you smell it?"

Marine microbes serve as an important source for commercial bioactive compounds. The present research is focussed on the Actinobacterial diversity of Machilipatnam coast. Actinobacteria are Gram positive bacteria that resemble Fungi in having filaments forming mycelia colonies. Owing to their morphological and cultural characteristics Actinobacteria are considered a group other than Bacteria. The different Actinobacterial Strains were studied having an ability to utilize the various carbon compounds as source of energy. 27 isolates of Actinobacteria including white, green, grey, orange and pink with different morphological types were isolated from Station I and II. Among them 19 isolates were from Pedapatnam and 27 from Polatitippa. The 27 identified species were falling under 10 genera including Actinobispora, Actinomadura, Actinomyces, Microbispora, Nocardis, Nocardiopsis, Saccharomonospora, Streptomyces, Streptosporangium and Thermomonospora. Streptomyces was the most dominant genus. For the evaluation of antibacterial activity, clinical strains of bacteria such as Gram positive Staphylococcus aureus, and Streptococcus faecalis and Gram - negative Proteus vulgaris and Salmonella typhi were used. Streptomyces alboniger, S.coelicolor and S.griseus were selected to study their antagonistic activity against the above mentioned clinical bacteria.

Petrichor

"Tch."

A drop of water landed on his nose, and Chuuya clicked his tongue. For a moment, he considered using his ability to get home quickly.

Too much hassle. It was his day off anyway. No use attracting attention.

Taking out the folding umbrella, he opened it, just as more water poured down from the sky, and resumed walking, the Krug Grande Cuvée case tucked neatly and carefully inside his coat.

People were moving around him. Some looking for shelter. Some running faster. Some taking out their umbrellas, all black and navy and dark colours, blended into the dark gray of the sky. Chuuya felt a little out of place, with his orange hair and burgundy umbrella.

Yokohama was quiet today.

He let his guard down a little bit, taking in the precious rare peacefulness, let instinct take over and feet carry him down the familiar way home. Passing the also familiar deserted park-

"What the fuck is he doing here!?"

Chuuya frantically looked around. Is Dazai here? Is anyone else here? Is he ambushed? Is he going to be shoved into another cursed book?

He wouldn't be fooled again. Not this time. Not by the same trick.

Two minutes passed, slow and torturous.

The detective just sat there, on the wooden bench, under the protection of his transparent umbrella. Alone.

Another two minutes passed.

Chuuya didn't know what made him do what he did, but he slowly took a step forward, and then another, and another, until...

The detective still sat there.

"Hey Mr. Fancy Hat." Ranpo's voice was soft, drowned out by the deafening rain, but Chuuya heard it all the same.

"...Edogawa."

"That sounds so old. Feels like I'm forty already. Just Ranpo!" He whined, just like a child, high pitch with a pout on his face, and Chuuya let out a small laugh.

"Fine, whatever. Ranpo."

Seemingly satisfied, Ranpo continued watching the rain, legs swinging occasionally, disturbed the puddle under his feet, humming a tune that Chuuya couldn't recognised. He vaguely recalled their time in the book, how the detective had run around, laughing and smiling childishly, eyes bright with excitement and uncontained joy, as if they hadn't been surrounded by five hundred dangerous and armed murderers. How his face had lighten up when saw a candy shop, then proceeded to drag Chuuya in there with him and conveniently identified a murderer while choosing lollipops.

"...- still here?"

"Ha?"

"I asked why you were still here. Wouldn't wanna keep that nice Krug Grande Cuvée and your Wagyu beef waiting, right?" How Ranpo knew about his plan for dinner, he wasn't even surprised anymore.

You look lonely.

"I do whatever I want." was what he said.

"Huh."

Chuuya took a look at Ranpo. Despite being protected by the transparent umbrella, the front of his white button was slightly damp, his white knee highs were completely soaked, the brown coat did little to hide his trembling shoulders. But he didn't seem bothered, instead tilting his head back a little bit more, as if to enjoy the rain easier.

Chuuya subconsciously moved closer, slightly tilted his own umbrella over the detective.

"How long have you been sitting here?"

"Until now, almost an hour."

"What the hell, why?" Is he crazy?

"I knew it was going to rain, so I came here to see if I timed it right. Of course this Great Detective is never wrong!"

"..."

The rain kept falling. Pitter-patter, pitter-patter. Deafening.

"...You like the rain. Why?" Chuuya dared to ask. Would Ranpo answer? Would he keep silence? Would he leave? Was this a mistake? So many possibilities, and Chuuya wasn't really a positive person. But as Ranpo opened those eyes, those damned clear, innocent, emerald eyes filled with nostalgia and melancholy, he held his breath, and waited.

One minute passed.

"Petrichor."

"...What?"

"Petrichor. That warm and fresh scent when rain falls on dry soil, stems from microscopic streptomycete bacteria in the soil that produce a compound called geosmin. You really need to improve your vocabulary, Fancy Hat-"

"I freaking know what petrichor is-!"

"I simply just like that scent."

Pitter-patter, pitter-patter.

"Why?"

"...When I was young, whenever it rained, Farther and I would sit outside, under the porch. He always let me sit on his lap, told me all about the cases he had solved, rubbed my arms to keep me warm." Ranpo unconsciously lifted his free hand and started rubbing his arm. "Mother would make tea and zenzai, then we would drink tea and eat zenzai together. They always let me have all the red beans..."

He had such a far away look in his eyes, as if he was a child once again, voice so small it was almost a whisper.

"I can hardly remember their faces now, you know. But somehow I remember that scent, petrichor." Ranpo shrugged. "So I thought: If I breathe deep enough, long enough, maybe I can keep them close in me, somehow."

Chuuya just stared at him, once again amazed by the detective, by his honesty.

By the amount of trust Ranpo put in him, enough to show him his real emotions, a moment of weakness.

They said nothing afterwards. Ranpo still sat there, and Chuuya just stand there, hoping that his umbrella was enough to keep Ranpo warm and dry.

Ten minutes passed. The rain stopped.

Ranpo stood up so sudden that Chuuya almost dropped his wine case. After closing their umbrellas, he turned the mafioso, a soft smile on his youthful face and in those innocent green eyes, and clutched his sleeve, not unlike the way a child would clutch an adult's pinkie.

"Let's go Fancy Hat! There's a candy shop around here! I want Ramune too!"

"Dammit Ranpo! I'm not your fucking bodygu-"

"That wine and steak has already waited for an hour and a half. What's another 30 minutes?"

Chuuya just smiled helplessly, and let Ranpo drag him away.

He took a deep breath.

Petrichor smelled really nice.

Prospects of Rare Actinomycetes for the Production of Newer Antibiotics- Juniper Publishers

Abstract

Rare actinomycetes are the genera of actinomycetes other than Streptomycetes found in less expored environment. Some of the actinomycetes genera such as Sinomonas, Nocardia, Micromonospora, Actip lanes, Pseudonocardia, Actinomadura, Micrbobispora are known as the novel species. Rare actinomycetes have been found to produce extremely useful secondary metabolites. They have the good antagonistic activity against the various human pathogens. Hence, it is believed to give a promising output of commercially important product for pharmaceutical industries.

Introduction

Actinomycetes are widely spread in nature, which are recognized to produce novel secondary metabolites. Among the overall genera in nature, actinomycetes by itself produce 70-80% of the existing antibiotics. In that, Streptomycetes found to produce 74% of antibiotics, which possess altogether 7,600 compounds lead of therapeutic agents like streptomycin, chloramphenicol, tetracycline, macrolides respectively. Production of novel drugs faces many problems, due to the rediscovery ofthe known secondary metabolites. It has driven the industrial attention towards the rare actinomycetes. Actinomycetes from the unexplored habitats seem to possess the potential antimicrobial activities against the human pathogens. The critical situation faced by various pathogens from hospitals and industries have lead to the demand for new drugs and chemical compounds in pharmacology. Hence, poorly explored area to be explored much for the novelty in the screening of rare actinomycetes. Actinomycetes are capable to grow in extreme environments like hot and dry deserts, hyper salinity, hay, sea water/sea muds, mangrove sediments, caves etc. Researchers are now focusing on the isolation of rare actinomycetes which are non- streptomyces. Genera such as Sinomonas humi sp. [1], Salinispora sp., Micromonospora, Actinoplane, Nocardia caishiienris and Pseudonocardia Carboxydivorans [2], Thermomonospora sp., Saccaropolyspora sp., Corynebacterium sp. [3] respectively are isolated as the novel stains.

Habitats of Rare Actinomycetes

Diversity of actinomycetes depends on the environmental microflora. Takahashi et al. describes that the diverse actinomy cetes found taxonomically and physiologically at desert soil [4]. He could isolate various genus like Microbispora, Nocardia, Microtetraspora/ Actinomadura, Amycolatopsis respectively from the Moava desert, California. Hamedi et al. represented novel species like Nocardia arvandica isolated from the sandy soil [5]. Nocardiopsis strains are found to grow in alkaline soils with high salt concentration at moderate temperature (37 °C). Nakaew et al. described that the genus Micromonospora isolated from Thai cave soil with low nutrient and high humidity, encourages the production of novel antibiotics and hydrolytic enzymes [6]. Phatup cave Forest park and Phanangkhoi cave in north Thailand were studied for the development of new bioactive compounds from rare actinomycetes. Endophytic actinomycetes are studied from the medicinal plants of Tropical rain forest in Xishuangbanna, China. Qin et al. explored the entophytic environment with the novel species like Saccaropolyspora, Dietzia, Blastococcus, Dactylosporangium, Promicromonospora, Oerskovia, Actinocorallia and Jiangella respectively [7].

Several interesting unusual habitats like the sediments of lakes [8], fungal cultivar [9] and ant workers produces antimicrobial products with the symbiotic association of actinomycete bacteria [10], Mangrove sediments [11], deep sea biodiversity [12], Marine sedimentation [13] respectively has reported the novel species of actinomycetes, with the pharmaceutical potent.

Antimicrobial Activity

Marine actinomycetes are found to have very good potential for the production of novel bioactive metabolites [14]. Secondary metabolites produced by the rare actinomycetes have potential antibacterial activity [15] has reported the Salinospora species from Fijian marine sponge, which showed the activity against the Methicillin-resistant Staphylococcus aureus stains. Qin et al. also reported that the species of rare actinomycetes like endophytic Pseudonocardia alni and Pseudonocardia zijingensis also showed significant antagonistic activity against the Methicillin-resistant Staphylococcus aureus stains [7]. Genus Micromonospora produces novel compounds like antraquinones and lupinacidins, which showed the anti-tumor activity. Tanvir et al. also found that the Pseudonocardia carboxydivorans exhibiting the anti-tumor activity [2].

Conclusion

Exploring the untapped area leads to the isolation of novel rare genera of actinomycetes with new bioactive compounds. Proliferation of new genera leads to the production of neobioactive compounds. Rare actinomycetes had increased the interest in tracking the microbial groups in the drug discovery process.

Marine Drugs, Vol. 15, Pages 286: Biodiversity of Actinobacteria from the South Pacific and the Assessment of Streptomyces Chemical Diversity with Metabolic Profiling

Recently, bioprospecting in underexplored habitats has gained enhanced focus, since new taxa of marine actinobacteria can be found, and thus possible new metabolites. Actinobacteria are in the foreground due to their versatile production of secondary metabolites that present various biological activities, such as antibacterials, antitumorals and antifungals. Chilean marine ecosystems remain largely unexplored and may represent an important source for the discovery of bioactive compounds. Various culture conditions to enrich the growth of this phylum were used and 232 bacterial strains were isolated. Comparative analysis of the 16S #rRNA gene sequences led to identifying genetic affiliations of 32 genera, belonging to 20 families. This study shows a remarkable culturable diversity of actinobacteria, associated to marine environments along Chile. Furthermore, 30 streptomycete strains were studied to establish their antibacterial activities against five model strains, Staphylococcus aureus, Listeria monocytogenes, Salmonella enterica, Escherichia coli and Pseudomonas aeruginosa, demonstrating abilities to inhibit bacterial growth of Gram-positive bacteria. To gain insight into their metabolic profiles, crude extracts were submitted to liquid chromatography-high resolution mass spectrometry (LC-HRMS) analysis to assess the selection of streptomycete strains with potentials of producing novel bioactive metabolites. The combined approach allowed for the identification of three streptomycete strains to pursue further investigations. Our Chilean marine actinobacterial culture collection represents an important resource for the bioprospection of novel marine actinomycetes and its metabolites, evidencing their potential as producers of natural bioproducts. http://bit.ly/2gWWQPI #MDPI
