Howl giving Sophie the ring in the movie has so much meaning. Howl, a self professed coward, someone who always runs away from his problems gives her a way to always find him/his heart. He is saying that he loves her and is basically giving her his heart.

When she thinks she might have killed them her heart calls out to Calcifer (his heart) just like Howl told her too and the magic of the ring leads her to exactly when and where she needs to be to save his heart.
